The Nexus
Within the Nexus, the MoL faction is made up out of those whose goals are to study the supernatural, further their knowledge & similar aspirations. They are also proficient alchemists and builders with a lot of knowledge about the other factions as well as monsters, creatures and (Nexus) history.
Origins
How, when and by who the Men of Letters were originally founded is unknown, but at some point in time, powerful humans banded together for the pursuit of knowledge and monster-hunting. Over time, the organisation branched out from wherever it started - it can now be found in a number of countries, including but not limited to the United States and the United Kingdom.

The crest of the Men of Letters is the Aquarian Star, a six-pointed sign that represents great magic and power and was linked to the gates of Atlantis.

The American Men of Letters organisation is known to have grown to both a significant number of members, and to great political power, as well as possessing powerful weapons and locations. They were a force to be reckoned with, and even claimed the glory of stirring the destruction of the Grand Coven by whipping the church into a frenzy, causing many of the Grand Coven's best members to be burned at the stake or forced into hiding.

The Men of Letters also know a great deal about magic, having stolen many of the Grand Coven's best spells and potions after its destruction. They also have knowledge of ancient and powerful magic of their own, including exorcism and time travel spells.

The Faction was lead by the Faction Elders, who made all the important decissions, and took care of all the orders business. Despite the name of the group, both women and men alike were allowed into the Order.

There was a bunker located in Kansas which held a great wealth of artifacts and knowledge, and was only accessible with a special key as it was a hidden base for the Men of Letters. Currently it is held by the Winchesters after the downfall and throughout the possible resurrection of the MoL faction in the United States. It has played an important role in the USA MoL history as well.
Downfall & Resurrection
In 1958, two initiates were sent out on an assignment, as part of their last test before becoming members of the order. Henry Winchester and Josie Sands were to investigate some events in the St. Bonaventure Convent. A nun had murdered two victims before she threw herself from a tower.

While Henry and Josie investigated, they learned that a group of demons were possessing the nuns, and even though they were able to exorcise a couple of them, the job turned out to be way too much for two initiates. This was because the demons weren't acting on their own accord but were instead serving one of the Knights of Hell, Abaddon.

Abaddon quickly overpowered Henry. She wanted to posess him so she could study the Men of Letters, but Josie begged the demon not to, and offered herself as a vessel instead. Abaddon agreed to this and ordered the rest of the demons to play dead so she could convince Henry that they successfully exorcized all the demons. Abaddon, while possessing Josie, also subtly threatened the surviving nuns to keep the ordeal quiet, thus preventing them from warning Henry of the truth.

On the 12th of August, 1958, Josie and Henry were to be initiated. Abaddon saw her chance during this event to attack. She destroyed most of the order, and followed Henry and the only surviving Elder into the year 2013 when the two MoL time-travelled, where she killed them too. With their deaths, The Men of Letters were extinct in The United States.

Later, Sam Winchester took up an interest in the MoL organisation and became the newest member of the previously extinct USA Men of Letters. Others showed an interest as well, and so the order has a potentially better future.
British Men of Letters
This large organisation, while mainly active in the UK, also has branches in other countries such as Spain, France, Germany, Ethiopia, and so on. Notably, their USA branch was wiped out.

The British MoL uphold a very strict code of conduct - anyone who fails to abide by the rules are killed instantly. One of the rules is that kiling a MoL without an order to do so is punishable by death; whether or not said death was accidental, done by a regular member or a legacy is irrelevant. If the deceased MoL member is not avenged, the person/MoL who failed to avenge them is executed instead. When it comes to monsters, their code is incredibly black and white: if something is a monster, harmless or not, it must always be killed.

This organisation is vast and very powerful. They have a widely placed system of wards and sigils in airports and docks, which alert them when monsters enter the country. Within 20 minutes, the monster is tracked and captured, and after 40 minutes, it’s dead. No exceptions. Thanks to this method of operating, there has not been a monster-related death in Britain since 1965.

Aside from this, the British Men of Letters are also highly capable in alchemy, having created devices by combining technology, craftsmanship and magic. With these weapons, which include but aren’t limited to a weapon capable of killing vampires through irradiation and hyperbolic pulse generators which can force an angel or demon from its vessel, they’ve forged themselves the ability to defeat monsters very efficiently.

The order had also been keeping tabs on Sam and Dean Winchester. After these two hunters found out about the British MoL’s plans to annihiliate american hunters and their families who wouldn’t cooperate with their plans, the american hunters led an assault on the British MoL branch in the USA and since then, the connection has been severed.
